Ganoderma spore oil promotes apoptosis of human lung adenocarcinoma SPC-A1 through downregulation of miR-21
OBJECTIVE: To investigate the effect of Ganoderma lucidum spore oil on proliferation, apoptosis and expression of miR-21 and its target genes in human lung adenocarcinoma cell line SPC-A1.

METHODS: The human lung adenocarcinoma cell line SPC-A1 was treated with Ganoderma lucidum spores oil for 24 h and 48 h. The cell proliferation was detected by CCK-8 method. The morphological changes were observed by inverted phase contrast microscope and flow cytometry Real-time PCR was used to detect the expression of miR-21 and its target genes PTEN and PDCD4. Results: The spore oil of Ganoderma lucidum could inhibit the proliferation of human lung adenocarcinoma cell line SPC-A1 in a time-dependent manner. When the volume fraction of Ganoderma lucidum spores reached 0.2%, the morphological changes of the cells were observed. Annexin-V / PI double- Ganoderma lucidum spores oil could promote the apoptosis of the cells. The spores oil of Ganoderma lucidum could down-regulate the expression of miR-21 and up-regulate the expression of PTEN and PDCD4 gene in SPC-A1 cells. Conclusion: Ganoderma lucidum spores oil can significantly inhibit the proliferation of human lung adenocarcinoma cell line SPC-A1 and induce cell morphological changes. Up-regulated the expression of PTEN and PDCD4 by down-regulating the expression of miR-21, so as to promote the apoptosis of lung cancer cells and achieve the anti-tumor effect.
